The victory handed the Brewers their first domestic cup title since 2016.
Striker Ian Simiyu opened the scoring for Tusker before Abud Omar restored parity for Kenya Police just two minutes later with a well-taken free-kick.
Simiyu, however, struck again in the 38th minute to complete his brace and restore Tusker's lead as the Brewers headed into the half-time break leading 2-1.
Kenya Police pushed hard for an equaliser in the second half and nearly found one in the 66th minute when Daniel Sakari unleashed a powerful long-range effort, but the Tusker goalkeeper produced a crucial save to preserve his side's advantage.
Tusker held firm in the closing stages to secure the trophy and cap off their season with silverware, ending a decade-long wait for the prestigious cup.
In the third-place playoff played earlier at the same venue, Bandari defeated KCB 2-0 courtesy of goals from Amza Mubarak and Andrew Juma.
The playoff preceded the final as part of the FKF Cup finals day programme in Kwale.
Tusker will now represent Kenya in the CAF Confederations Cup.
Police defender Aboud Omar was named the defender of the tournament; Ian Simiyu from Tusker bagged the double award: midfielder of the tournament and most valuable player of the tournament.
In addition, Humphrey Aroko and Amza Mubarak both bagged top scorer of the tournament with six goals apiece. Elvis Ochoro from KCB went home with best goalkeeper of the tournament.
